Anglade was previously a member of the Coalition Avenir Québec. In the 2012 election, she ran as the CAQ candidate in Fabre, losing to Liberal Gilles Ouimet. She served as the CAQ's president from 2012 to 2013. She resigned in order to become the CEO of Montreal International.

In 2015, Anglade joined the Quebec Liberal Party and ran as their candidate for Saint-Henri–Sainte-Anne in a by-election. She cited objections to the CAQ's stances on ethnic identity and immigration to explain her political turn.

From 2016 until 2018, Anglade served as Minister of Economic Development, Innovation, and Export Trade in Philippe Couillard's administration. This made Anglade the first Canadian of Haitian ancestry to hold a ministerial position. In 2017, Anglade was appointed Deputy Premier of Quebec, a position she held until the collapse of the Liberal government in the 2018 election.

On May 11, 2020, Anglade was appointed party leader after her opponent, Alexandre Cusson (Fr), withdrew from the campaign.She became the first woman to lead the Quebec Liberal Party and the first black woman in Quebec to lead a provincial party.On November 7, 2022, Anglade announced her retirement as leader of the Quebec Liberal Party.
