Gakpo made his debut with the PSV first team on February 25, 2018, coming on as a substitute in the final minute of injury time in a 3–1 win over Feyenoord.During the 2018–19 season, Gakpo was able to demonstrate even greater improvement in his overall play. 

On December 3, 2018, he earned the first hat trick of his professional career in the home win over the Go Ahead Eagles that ended with a score of 5-1.

He made his debut in the Eredivisie just one day after scoring a goal and assisting on another during the team's 5–2 victory at home over Almere City on the 21st of December . 

On February 3, 2019, he made his second appearance in the league and scored his first goal during a 5–0 home victory over Fortuna Sittard. He also assisted on another goal during the game.By the time the season was out, he had played in the league on 14 different occasions, but he was unable to score any more goals.

In the 2019–20 season, he made his debut in the Eredivisie and went on to collect seven goals and the same number of assists in a total of 25 league contests.

Gakpo scored his first brace for PSV on the first matchday of the 2020–21 Eredivisie season, which was played on September 13, 2020. His contributions helped PSV earn a 3–1 victory over Groningen.

PSV's victory over Ajax in the 2022 KNVB Cup Final came courtesy of a goal scored by Gakpo. PSV's final score was 2–1.

On August 31, 2022, he recorded his first hat-trick in a victory over Volendam that ended with a score of 7–1.

Gakpo reportedly drew interest from Manchester United, Leeds United, and Southampton during the summer transfer window of 2022; nevertheless, he opted to remain with PSV for the ensuing 2022–2023 campaign.
