From 1999 to 2001, Chalmers worked as a research officer in Queensland Premier Peter Beattie's Department of Premier and Cabinet. From 2002 to 2004, he was the ALP's national research manager; from 2005 to 2006, he was Shadow Treasurer Wayne Swan's media adviser; in 2006, he was Opposition Leader Kim Beazley's deputy chief of staff; and from 2006 to 2007, he was a senior adviser to New South Wales Premier Morris Iemma.

Chalmers returned to work for Wayne Swan in the Treasury after Labor won the federal election in 2007, first as deputy chief of staff and principal adviser (2007-2010) and then as chief of staff (2007-2010). (2010–2013). He briefly served as executive director of the Chifley Research Centre in 2013. In the same year, he published Glory Daze, a book about the disparity between Australia's stellar economic performance and public discontent with the government.

Chalmers was elected to parliament as a member of Labor Right in the 2013 federal election, replacing retiring ALP member Craig Emerson in the Rankin Division. He defeated former MP Brett Raguse for Labor preselection. Chalmers became a shadow parliamentary secretary in October 2013, a shadow minister in October 2015, and the Shadow Minister for Finance following the 2016 election. In 2016, he co-founded the Courtyard Group, a roundtable of Labor MPs and "Australia's greatest progressive thinkers from industry, academia, media, and consumer advocacy organizations."

Following the 2019 federal election, Chalmers expressed interest in taking over as party leader and Leader of the Opposition from Bill Shorten. His youth and status as a Queenslander, as well as his membership in the Labor Right faction, were viewed as potential assets. Several members of his party, however, had already decided to vote for Anthony Albanese of the Labor Left. Finally, Chalmers declined to run for the seat, allowing Albanese to take over unopposed. He then ruled out running for vice president as well. He was later named Shadow Treasurer in Albanese's administration.
